چکیده

It is known that the requirement of microorganisms for certain growth factors is influenced by the nutritive medium in a manner analogous to the effect of dietary regimen of animals on their vitamin requirements. In this connection it was found by Williams and Fieger (1) that oleic acid could substitute for biotin in the growth of Lactobacillus casei under their experimental conditions. Axelrod et al. (2) have recently shown that this is also the case for Lactobacillus arabinosus. Koser et al. (3) were able to demonstrate that, for To&a cremoris, aspartic acid could largely substitute for biotin. More recently Stokes, Larsen, and Gunness (4) have shown a definite relationship between biotin and aspartic acid and were able to demonstrate that aspartic acid synthesis resulted when biotin was present, Since Williams and Fieger (1) demonstrated that oleic acid does not cause biotin synthesis, it appears possible that biotin is responsible for synthesis of both aspartic acid and oleic acid. The purpose of the investigation herein reported is to elucidate the relationship between biotin and these two nutrilites and to determine whether there is a relationship between aspartic acid and oleic acid other than their common connection with biotin.

Factors affecting the accumulation of biotin by Lactobacillus arabinosus.

The problem of substrate accumulation in microbial cells has been the subject of considerable investigation in recent years. However, comparatively little is known of the conditions influencing the accumulation of vitamins into microbial cells. One might cite as examples the investigations of Oginsky (1952) with vitamin B12, of Lichstein and Ferguson (1958) with biotin, and of Wood and Hitching...

Microbiological Activity of Synthetic Biotin, Its Optical Isomers, and Related Compounds

In a previous report, synthetic and natural biotin were stated to have identical growth-promoting properties for Lactobacillus arabinosus 17-5 (1). This paper gives a more detailed comparison of these substances in the growth of four other biotin-requiring microorganisms. The activities of synthetic dland I-biotin, dl-allobiotin, dl-diamino acid sulfate, desthiobiotin,’ and the probable biologi...

The influence of biotin nutrition on carbon dioxide fixation and malic enzyme formation by Lactobacillus arabinosus.

Biotin has long been impliratcld in a number of carbon dioxide fixation reactions (see Lardy and Peanasky (1) for rcvicw). Two main proposals have been made to explain its mode of action: 1. The vitamin acts as a coenzyme in carbon dioxide fixation reactions. This view was based mainly on observations on the incorporation of radioactivity from C?“oz into compounds in equilibrium with certain su...

Cell permeability: a factor in the biotin-oleate relationship in Lactobacillus arabinosus.

Bound biotin-saturated cells were incubated in the presence of biotin and glucose (37 C, pH 7.5) with or without oleic acid, Tween 20, 40, 60, and 80, Aerosol OT, sodium dodecyl sulfate (SDS), cetyltrimethylammonium bromide, Triton X-100, Non-Ion-Ox, and Haemo-Sol. With low concentrations (up to 5 mug/ml) and short reaction times (up to 10 min), oleic acid stimulated free biotin accumulation. I...

The indirect role of biotin in the synthesis of the malic enzyme.

Although biotin is required for the synthesis of the inducible malic enzyme in Lactobacillus arabinosus 17-5 (I), partially purified preparations of the enzyme do not contain biotin (a), and avidin does not affect the activity of the enzyme (3). The formation of another enzyme, ornithine transcarbamylase, in iStreptococcus Zactis 8039 is also diminished by a biotin deficiency (4), but purified ...
